Mark 70 mm above the extruder, extrude 50 mm slowly, and a calibrated extruder leaves 20 mm.

Heat and load

Load filament. Heat the hotend to its printing temperature.

Mark and measure

Mark the filament about 70 mm above the extruder intake. Measure from the extruder body to the mark with calipers.

Extrude 50 mm slowly

Send G91, then G1 E50 F60, and wait. It takes about 50 seconds, and the slow rate matters, so skip the extrude button on the printer's screen or in a web interface, which pushes filament far faster.

Measure again

Measure from the extruder body to the mark a second time. The difference between the two readings is what it really extruded.

Klipper's guide warns that a fast rate builds pressure in the extruder and skews the result. At 60 mm per minute the hotend keeps up.
On Marlin, send M503 and read the E value on the M92 line. On Klipper, it is rotation_distance in the [extruder] section of printer.cfg.
E-steps first. It fixes how much filament the extruder pushes. Flow rate then fine-tunes how much each filament needs.
